胃痞消治疗慢性萎缩性胃炎大鼠的疗效观察及机制探讨

目的:探讨健脾祛瘀解毒中药复方胃痞消对慢性萎缩性胃炎(chronic atrophic gastritis,CAG)的疗效及机制。方法:将SD大鼠随机分为正常对照组、模型对照组、胃痞消组、胃复春组。除正常对照组外,其他组均采用MNNG(N-甲基-N′-硝基-N-亚硝基胍)饮水液自由饮用、饥饱失常、40%乙醇和雷尼替丁溶液(3mg/ml)交替灌胃法复制CAG大鼠模型,连续8周。从第9周起,给药组按照上述剂量连续灌胃给药共12周,从胃液分泌、血清学、病理组织学角度观察胃痞消对大鼠胃黏膜影响。结果:与正常组相比,模型组大鼠胃液酸度、胃蛋白酶活性、胃肠激素含量均显著降低(P<0.01),黏膜层显著变薄、黏膜肌层显著变厚、腺体显著减少(P<0.01);与模型组相比,胃痞消组、胃复春组大鼠胃液酸度、胃蛋白酶活性、胃肠激素含量均有不同程度增加(P<0.01,P<0.05),大鼠胃壁黏膜层变厚,黏膜肌层变薄,腺体增加(P<0.05);与胃复春组相比,胃痞消组大鼠胃液酸度、胃蛋白酶活性、胃肠激素含量均有增加趋势,但无统计学意义,胃痞消组黏膜层变厚,黏膜肌层变薄,腺体增加,亦无统计学意义。结论:胃痞消对CAG大鼠模型具有良好的治疗作用,可以促进胃液分泌,提高胃液酸度,增加胃蛋白酶的活性,减轻腺体萎缩。

Efficacy and Mechanism of Weipixiao in Treating Rats with Chronic Atropic Gastritis

Objective: To study the effect of strengthening spleen,activating blood,and detoxification Chinese Herbal Compound Weipixiao in chronic atrophic gastritis(CAG) rats. Methods: SD rats were randomly divided into normal group,model group,Weipixiao group(20 g/kg.d),Weifuchun group(0.4 g/kg.d),except for the normal group,the rats in other groups were fed with 160μg/ml of MNNG water,irregular diet,lavage 40% ethanol and Ranitidine’s(3 mg/ml) solution to establish model rats of CAG for 8 consecutive weeks. At the beginning of 9 th week,the mediation group were provided with treatment for 4 weeks,and observing curative effects of Weipixiao on gastric mucosa in rats from the point of gastric juice secretion,serology and histopathology. Results: Compared with those in the normal group,in the model group,gastric acidity,pepsin activity and gastrointestinal hormone content were significantly decreased(P<0.01). The mucosal layer was thinner evidently,the mucosal muscular layer was thickener remarkablely,and the glands were decreased significantly(P<0.01). Compared with the model group,in the Weipixiao group and the Weifuchun group,gastric acidity,pepsin activity and gastrointestinal hormone content were increased variously(P<0.01,P<0.05),mucosal layer was thickener,mucosal muscular layer was thinner,and glands were increased(P<0.05). Compared with the Weifuchun group,the gastric acidity,pepsin activity and gastrointestinal hormone content in the Weipixiao group were increased,but the difference not being significant,mucosal layer was thickener,mucosal muscular layer was thinner,and glands were increased,the difference between them not being significant either. Conclusion: Weipixiao has a greatly curative effect on CAG rats model,Weipixiao can promote the secretion of gastric juice,increase the acidity of gastric juice,increase the activity of pepsin,and reduce the atrophy of glands.
