香港远志提取物对局灶性脑缺血大鼠脑保护作用与作用机制

摘    要:目的探讨香港远志提取物对局灶性脑缺血大鼠脑保护作用与机制。方法线栓法制备SD大鼠大脑中动脉闭塞(MCAO)模型,随机分4组:假手术组、模型组及治疗A、B组。治疗组按设定方式给药,假手术组、模型组给1%吐温溶液。观察术后24h神经功能缺损评分(NBDS)、脑梗死体积(IV)、血清神经元烯醇化酶(NSE)及神经元凋亡、Bcl-2、Bax表达。结果与假手术组比较,模型组和治疗组NBDS、IV、NSE、神经元凋亡及Bcl-2、Bax显著增高、Bcl-2/Bax显著降低(P<0.05)。与模型组比较,治疗组NBDS、IV、NSE及神经元凋亡、Bax显著降低、Bcl-2、Bcl-2/Bax显著增高(P<0.05),治疗组间数据亦有显著差异(P<0.05)。结论香港远志提取物预处理,对局灶性脑缺血大鼠脑神经有保护作用,机制可能是促进Bcl-2、抑制Bax表达,上调Bcl-2/Bax比值,阻止神经元凋亡。

Neuroprotective effect and mechanism of polygala hongkongensis hemsl extracts on the focal cerebral ischemia in rats

Abstract:Objective To investigate the neuroprotective effect and mechanism of polygala hongkongensis hemsl extracts(PHHE)on the focal cerebral ischemia in rats.Methods SD rats were randomly divided into 4 groups,sham operated group,model group and treatment group A and B.Treatment groups received drug respectively according to grouping protocol,the sham operated group and model group received 1% tween-80 solution.The middle cerebral artery occlusion(MCAO)was made by thread approach,at 24h after MCAO,the neurological behavior deficits scores(NBDS),infarction volume(IV),NSE and neuronal apoptosis and expressions of Bcl-2 and Bax were observed.Results Compared with sham operated group,the NBDS,IV,NSE,neuronal apoptosis and expressions of Bcl-2 and Bax were significantly increased and the Bcl-2/Bax was significantly decreased in both the model and treatment groups(P<0.05).Compared with model group,the NBDS,IV,NSE,neuronal apoptosis and Bax were significantly decreased and Bcl-2 and Bcl-2/Bax were significantly increased in treatment groups(P<0.05).The data between treatment groups showed significantly difference(P<0.05).Conclusions PHHE given prior to the focal cerebral ischemia plays a neuroprotective role which may be achieved by increasing the expression of Bcl-2,decreasing that of Bax,increasing Bcl-2/Bax and inhibiting neuronal apoptosis.
