硝酸甘油偏头痛模型对大脑即早基因表达的影响及盐酸氟桂利嗪的干预作用

摘    要:目的探讨硝酸甘油偏头痛模型对脑即早基因表达的影响及盐酸氟桂利嗪的干预作用。方法健康Wistar大鼠30只,随机分为对照组、偏头痛模型组、治疗组,各组均灌胃给予盐酸氟桂利嗪或等量生理盐水,每日1次,连续7 d,末次给药20 min后,皮下注射生理盐水或硝酸甘油复制实验性偏头痛大鼠模型,于3 h后取材。随机选取大鼠脑片5个视野,进行c-fos和c-jun阳性细胞计数,分析每张切片上每个视野的平均灰度。结果偏头痛模型组脑组织切片c-fos和c-jun阳性细胞数(30.60±3.78)、(26.50±6.56)]较对照组(9.10±1.66)、(8.02±2.05)]明显增多(P〈0.01),盐酸氟桂利嗪治疗组c-fos和c-jun阳性细胞数(13.40±3.45)、(14.30±2.88)]较模型组显著减少(P〈0.05);偏头痛模型组脑组织切片c-fos和c-jun阳性细胞灰度(104.50±13.73)、(84.34±16.92)]较对照组(157.05±37.16)、(177.65±34.36)]显著减弱(P〈0.01);盐酸氟桂利嗪治疗组c-fos和c-jun阳性细胞灰度(129.18±37.2)、(151.44±43.29)]较模型组明显增强(P〈0.05)。结论硝酸甘油偏头痛动物模型可显著提高c-fos和c-jun阳性细胞数,减弱c-fos和c-jun阳性细胞灰度,而盐酸氟桂利嗪可抑制实验性偏头痛动物模型脑内c-fos和c-jun的阳性表达,增强c-fos和c-jun阳性细胞灰度。

Abstract:Objective To investigate effect of Nitroglycerin migraine model on brain immediately early gene expression and intervention effect of Flunarizine Hydrochloride.Methods Totally 30 healthy Wistar rats were randomly divided into three groups: the control group,the Nitroglycerin migraine model group and the Flunarizine Hydrochloride treat ment group.Rats received Flunarizine Hydrochloride or normal saline by intragastric administration,once daily,for 7 successive days.20 minutes after last giving medicine,normal saline or Nitroglycerin were injected hypodermically to duplicate migraine model,and drew the materials 3 hours later.Randomly selected 5 horizons of the rat brain slices,and the positive cells were counted and average gray scale of each slice per field was analyzed.Results c-fos and cjun positive cells of migraine animal model group (30.60 ±3.78),(26.50±6.56)] increased significantly,compared with the control group (9.10±1.66),(8.02±2.05)](P〈0.01),c-fos and c-jun positive cells of Flunarizine Hydrochloride treatment group (13.40±3.45),(14.30±2.88)] notably decreased than the migraine animal model group(P〈0.05).Average gray scale of c-fos and c-jun positive cells of migraine animal model (104.50 ±13.73),(84.34±16.92)] significantly reduced than the control group (157.05 ±37.16),(177.65±34.36)](P〈0.01),that of Flunarizine Hydrochloride treatment group (129.18±37.2),(151.44±43.29)] augmented significantly compared with the migraine animal model(P〈0.05) in brain.Conclusion Nitroglycerin migraine animal model can significantly increase the positive cells number of c-fos and c-jun,and weaken positive cells average gray scale of c-fos and c-jun.Flunarizine Hydrochloride can inhibit brain c-fos and c-jun positive expression of experimental animal model of migraine,and enhance positive cells average gray scale of c-fos and c-jun.
