| 摘 要： | 目的:制备人源IgG抗-Di a试剂血清,调查乌鲁木齐地区献血者人群中Di a抗原分布频率。方法:用吸收试验方法制备IgG抗-Di a,用间接抗人球方法对987例无血缘关系的献血者进行Di a抗原的筛选,对含有Di a抗原个体用PCR-SSP方法进行基因分型。结果:既去除了B型血清中的抗-A,又未使IgG抗-Di a效价降低,检测效价及特异性符合IgG抗-Di a试剂血清要求,Di a抗原在本地区献血人群中的基因频率为0.011 1。结论:采用吸收方法,解决了人源抗-Di a试剂血清购置困难,同时获得了本地区献血人群中Di a抗原基因频率分布,为临床输血安全提供了保障。

| Abstract: | Objective： To preparate anti-Dia reagent serum, and investigate the distribution of Dia antigen in blood donors of Urumqi. Method：Anti-Dia reagent serum was preparated by absorption test,Dia antigen 987 sam- ples from unrelated blood donors in Urumqi were detected with indirect antiglobulin test fIAT）, and samples of Dia antigen were genotyped with Sequence-Specific Primers（PCR-SSP）. Result： The acquired reagent sera had a high anti-Dia titer with a high specificity, after anti-A was absorbed, and Dia gene frequency was 0. 011 1. Conclusion： Absorption test could be helpful to the production of hunman ongin anti-Dia reagent serum, moreover, the local Dia gene frequency distribution in our study would provide the guarantee for the safety of clinical blood transfusion. |
