肠祺饮对肝郁脾虚腹泻型肠易激综合征大鼠脑肠肽的影响

摘    要:目的观察中药肠祺饮对肝郁脾虚腹泻型肠易激综合征(IBS-D)大鼠部分脑肠肽的调控作用。方法 SPF级雄性Wistar大鼠72只,分为正常组、模型组、得舒特组和肠祺饮组,每组18只。除正常组外,其余3组均采用慢性束缚应激与灌服番泻叶煎剂相结合的方法建立具有肝郁脾虚证表现的IBS-D大鼠模型,分别给予0.9%Na Cl溶液、得舒特、肠祺饮水煎剂。观察大鼠血清5-羟色胺(5-HT)和下丘脑、结肠组织中P物质(SP)、血管活性肠肽(VIP)mRNA表达量变化。结果肠祺饮能够降低血清5-HT含量,并减少下丘脑和结肠SP mRNA、VIP mRNA表达(P0.05)。结论肠祺饮可能通过降低血清5-HT含量和减少下丘脑与结肠中SP mRNA、VIP mRNA表达,调节肝郁脾虚IBS-D大鼠脑-肠轴功能紊乱。

Abstract:  Objective:To observe the regulation effects of Changqi Drink on the levels or gene expressions of some brain-gut peptides in diarrhea-predominant irritable bowel syndrome(IBS-D) rats with syndrome of liver stagnation and spleen deficiency. Methods72 SPF male Wistar rats were randomly divided into the normal group,model group,Dicetel group and Changqi Drink group,18 rats in each group. Except the normal group,the IBS-D model with syndrome of liver stagnation and spleen deficiency was established by chronic restraint stress and folium sennae decoction in the other groups. Each group was treated with 0.9%NaCl solution,Dicetel and Changqi Drink decoction respectively. The serum level of 5-hoxytryptamine(5-HT) and the mRNA expressions of substance P(SP) and vasoactive intestinal peptide(VIP) in hypothalamus and colon tissue were detected.  Results:Changqi Drink could decrease the serum level of 5-HT and down-regulate the mRNA expressions of SP and VIP in hypothalamus and colon tissue(P<0.05).  Conclusion:Changqi Drink can regulate the dysfunction of brain-intestinal axis of IBS-D rats with syndrome of liver stagnation and spleen deficiency by decreasing the serum level of 5-HT and down-regulating the mRNA expressions of SP and VIP in hypothalamus and colon tissue.
