针刺任督脉对脑缺血再灌注大鼠血管内皮生长因子和CD34表达的影响

摘    要:目的 探讨任督脉经穴针刺对脑缺血再灌注大鼠脑内血管内皮生长因子(VEGF)和CD34蛋白表达的影响.方法 将雄性SD大鼠随机分为假手术组、模型组和针刺任督脉组,每组又随机分为7、14 d和28 d各三个亚组.线栓法制作大鼠大脑中动脉栓塞(MCAO)模型.假手术组切开后不插线直接缝合.利用免疫组化法检测7、14 d和28 d大鼠脑内VEGF和CD34蛋白的表达情况.结果 大鼠MCAO后模型组和针刺任督脉组VEGF和CD34蛋白表达均增加,其中,模型组VEGF表达在7d为最低(169.97±2.42,P<0.01),后逐渐增多,CD34表达在14 d为高峰期(164.17±2.86,P< 0.01),后逐渐减少.针刺任督脉组跟模型组的7、14 d和28 d相比,VEGF表达均增加(14.47±0.32) (P< 0.01)、(10.16±0.25)(P<0.01)、(9.17±0.21)(P<0.01),且CD34表达均增加(16.00±0.20) (P< 0.01)、(19.34±0.38) (P< 0.01)、(16.50±0.27) (P< 0.01).结论 针刺任督脉经穴可能通过上调脑内VEGF和CD34蛋白表达,促进微血管新生,这可能为其抗脑缺血再灌注损伤的作用机制之一.

Influence of conception and governor vessel acupuncture on vascular endothelial growth factor and CD34 protein expressions in rats with cerebral ischemia-reperfusion injury

Abstract:Objective To explore the influence of conception and governor vessel acupuncture on the vascular endothelial growth factor (VEGF) and CD34 protein expressions in rats with cerebral ischemia-reperfusion injury.Methods Male SD rats were randomized into sham surgery group,model group and acupuncture group,each again were randomized into 7-day,14-day and 28-day groups,respectively.With the thread embolism method,the models of middle cerebral artery occlusion (MCAO) were established,and the sham surgery models were made without thread insertion.The VEGF and CD 34 protein expressions in the brains were detected with immunohistochemistry.Results After modeling,the VEGF and CD 34 protein expressions increased.The VEGF expression was the highest on day 7 (169.97±2.42,P < 0.01) and then decreased,and the CD 34 expression was the lowest on day 14 (164.17±2.86,P <0.01) and then increased.Compared to model group in 7,14 and 28 d,the VEGF expression in the acupuncture group increased of (14.47 ± 0.32) (P < 0.01),(10.16±0.25) (P < 0.01),(9.17±0.21) (P < 0.01) respectively,and the expression of CD34 increased of (16.00 ± 0.20) (P < 0.01),(19.34 ± 0.38) (P < 0.01),(16.50 ± 0.27) (P < 0.01) respectively.Conclusion Conception and governor vessel acupuncture might promote angiogenesis via up-regulating the VEGF and CD 34 protein expressions,which may be one of the mechanisms for inhibiting the cerebral ischemia-reperfusion injury.
