天花粉蛋白诱导人乳腺癌MDA-MB-231细胞凋亡机制的研究

摘    要:目的:探讨天花粉蛋白( TCS)诱导人乳腺癌MDA-MB-231细胞凋亡及与凋亡相关蛋白bcl-2表达相关性。方法利用CCK-8检测不同浓度TCS对MDA -MB-231细胞生长抑制作用;通过流式细胞仪( FCM )测定天花粉蛋白作用后MDA-MB-231细胞周期及细胞凋亡变化情况;利用流式细胞仪观察不同浓度天花粉蛋白作用于MDA-MB-231后bcl-2蛋白表达情况。结果随着TCS浓度升高、作用时间延长,人乳腺癌MDA-MB-231细胞的生长受到明显抑制,且存在剂量效应和时间效应关系;TCS在一定浓度范围内能诱导人乳腺癌MDA-MB-231细胞凋亡,使细胞阻滞于G1期,并使细胞中bcl-2蛋白表达下降。结论 TCS能通过促进细胞凋亡来抑制人乳腺癌MDA-MB-231的生长,bcl-2表达下降可能是TCS诱导人乳腺癌MDA-MB-231凋亡的分子机制之一。

Abstract:Objective To reseach how do trichosanthin induce human breast cancer MDA-MB-231 cell apoptosis and observe the relationship between the expression of bcl-2 protein expression and TCS .Methods CCK-8 was mainly used to detect proliferation inhibition of MDA-MB-231cells for different concentrations of trichosanthin;FCM was used to detect the cell line and the apoptosis .Use FCM to detect the bcl-2 protein expression .Results With the TCS concentration increased,and the reaction time prolonged ,the proliferation inhibition of human breast cancer MDA-MB-231 cell was increased ,and therewere dose-response and time-effect relationship.When TCS in a certain range of concentration,it could induce human breast cancer MDA-MB-231 cells staged in G1 phase,in the same time it could reduce the expression of bcl-2 protein.Conclusion TCS can inhibite the growth of human breast cancer MDA-MB-231 due to the promotion of apop-tosis,and the mechanism of apoptosis maybe due to the induction in the expression of the bcl -2.
