金属螯合层析介质的研制及其在纯化重组人铜锌超氧化物歧化酶中的应用

摘    要:以琼脂糖Sepharose 4B为基质,经氯代环氧丙烷活化,与亚氨基二乙酸通过化学交联再与铜螯合,合成了金属铜螯合层析介质,结果显示此介质不仅具有特异性吸附能力,且有一定的刚性,对强碱和热亦有一定的耐受性.用此介质纯化人超氧化物歧化酶(rhSOD),即可得到高纯度的rhSOD产品,简化了纯化工艺,降低了生产成本.

Abstract:Sepharose 4B was used as substrate for chromatography, epichlorohydrin as activator and iminodiacetic acid as ligands ,then Cu was chelated to form Cu-metal chelate affinity chromatographymedium. The results showed that the medium not only had a specific adsorbent capacity to rhSOD ,butalso possessed rigidity in some degree and that its stabilities against alkali and heat were increased significantly. By using the medium,the rhSOD with high purity was obtained,the working processes were simplified and the production costs were reduced.
