变应性鼻炎患者外周血淋巴细胞蛋白激酶C活性变化及意义

摘    要:目的研究变应性鼻炎患者外周血淋巴细胞蛋白激酶C(PKC)活性变化及其与Th2细胞因子的关系。方法变应性鼻炎组和正常对照组外周静脉血中分离和纯化淋巴细胞胞质及胞膜PKC,采用同位素γ-32P-ATP催化活性测定法检测其活性,采用酶联免疫吸附试验(ELISA)法检测血清中细胞因子白细胞介素(IL)-4和IL-5在的含量。结果变应性鼻炎患者淋巴细胞PKC的总活性较对照组明显增强,且变应性鼻炎组胞膜PKC的活性及胞膜PKC活性占总活性的百分比值与正常对照组比较明显增高,且差异有统计学意义(P<0.05);变应性鼻炎组患者血清IL-4和IL-5水平显著高于正常对照组。结论应性鼻炎患者外周血淋巴细胞PKC的激活可能与炎症介质的释放增加有关;提示PKC通道可能参与T淋巴细胞的活化过程,PKC信号转导途径在变应性鼻炎患者的发病机制中可能具有一定的意义。

Abstract:Objective To investigate the changes of protein kinase C (PKC) activity in peripheral blood lym- phocytes (PBL) and the correlation with Th2 cytokines in patients with allergic rhinitis. Methods PKC obtained from the cytoplasm and cellular membrane of PBLs in patients with allergic rhinitis and normal controls was isolated and purified for activity assaying by isotope γ-P-ATP catalytic activity test and measurement of serum IL-4 and IL-5 by enzyme-linked immunosorbent assay. Results Patients with allergic rhinitis were associated with markedly higher PKC activity in lymphocytes as well as higher cellular membrane PKC activity and its ratio to the total PKC activity than normal controls (all P〈0.05). Additionally, higher levels of IL-4 and IL-5 were noted in patients with allergic rhinitis (both P〈0.05). Conclusion When compared with normal controls, patients with allergic rhinitis are featured by considerably increased total activity and the level of activation of PKC in PBL, a consequence that might be asso- ciated with release of inflammatory mediators, suggesting that PKC signaling pathway might be involved in the activa- tion of T lymphocytes and plays a crucial role in the pathogenesis of allergic rhinitis.
