热休克蛋白90抑制剂对视网膜色素上皮细胞增生的抑制作用

摘    要:目的 :探讨热休克蛋白 90 (heatshockprotein 90 ,Hsp90 )抑制剂对培养的视网膜色素上皮 (Retinalpigmentepithelial,RPE)细胞端粒酶活性以及RPE细胞增生的影响。方法 :用分子原位杂交方法分别检测对照组和Hsp90抑制剂组RPE细胞端粒酶活性 ;末端缺口标记 (terminalUTPnick endlabeling ,TUNEL)法分别检测上述 2组细胞中凋亡细胞的比率 ;在上述 2组细胞中分别用甲基偶氮唑蓝 (metheltetrazolium ,MTT)法检测细胞增生状况。结果 :与RPE细胞对照组相比 ,加入Hsp90抑制剂组RPE细胞端粒酶活性显著降低 ,凋亡细胞的比率明显增高 ,RPE细胞生长抑制率升高。结论 :Hsp90抑制剂可抑制RPE细胞中端粒酶的活性 ,从而抑制RPE细胞的增生并引起RPE细胞的凋亡

Abstract:Objective: To discuss the effect of heat shock protein 90(Hsp90) inhibitor on cultured retinal pigment epithelial(RPE) cell. Methods: The activation of telomerase on cultured RPE cell and RPE cell which co cultured with inhibitor of Hsp90 by molecule in situ hybrilization was measured;The rate of opoptosis cell in above two groups,and the proliferative status of RPE cells in above two groups by MTT method were also measured. Results: Compared with the group without inhibitor of Hsp90,the group with inhibitor of Hsp90 (Geldanamycin) was that the activation of telomerase decreased remarkably,and the rate of opoptosis cell increased remarkably,and that with increase of the concentration of inhibitor,the alive RPE cells decreased. Conclusion: The inhibitor of Hsp90 can restrain the activation of telomerase and the proliferation of cultured RPE cells,and then induce the opoptosis of RPE cells. This can provide a new method of treating PVR.
