摘    要:与一般植物叶片相比,果树叶片因富含多酚和多糖类物质,制备高分子量DNA比较困难。我们建立了一个个从果树叶片制备高分子量DNA的有效程序,按照该方法制备的五种果树HMWDNA,无叶绿体和线粒体DNA污染,无大量的非特异小片段的混杂,有利于限制性酶切分析和大片段基因组文库的构建。

Abstract:Compared with general plant leaves, fruit tree leaves are rich in polyphenols and polysaccharides. Therefore it is very difficult to prepare high molecular weight DNA from them. We developed an efficient high molecular weight (HMW) genomic DNA preparation procedure for fruit tree leaves. We successfully isolated HMW DNA from 5 species fruit tree leaves that represented the material background of fruit tree leaves. The HMW DNAs were with no chloroplast and mitochondrail DNA and little small molecular weight DNA contamination. It demonstrated that the HMW DNA was suitable for reproducible digestion by restriction enzymes, large genomic library construction and analysis of complex genome region.
