藏药韦色尼阿丸预防宫腔粘连形成的实验研究

摘    要:目的探讨藏药韦色尼阿丸预防宫腔粘连的效果及对子宫内膜中转化生长因子-β(TGF-β)和基质金属蛋白酶-9(MMP-9)表达的影响。方法选择6~8周龄、体重180~200 g的雌性未孕SD大鼠,随机分为4组:正常对照组、模型对照组、模型组、研究组,其中正常对照组未进行任何处理,模型对照组切开子宫后未进行造模,模型组采用机械损伤及酒精灼烧法进行造模,而研究组在造模后用藏药韦色尼阿丸灌胃处理14 d。利用HE染色观察比较各组大鼠子宫内膜形态及子宫内膜腺体数量变化;利用Masson染色比较子宫内膜纤维化面积;通过免疫组织化学染色法观察藏药韦色尼阿丸对TGF-β和MMP-9表达水平的影响。结果(1)研究组相对于模型组内膜厚度增加、腺体数目增多(P<0.05),而与正常对照组比较子宫内膜厚度和腺体数目均无显著变化(P>0.05);(2)研究组子宫内膜纤维化面积较模型组显著减少(P<0.05),而相对于对照组无显著变化(P>0.05);(3)研究组大鼠子宫内膜中TGF-β表达水平较模型组显著降低(P<0.05),而与对照组无显著差异(P>0.05);(4)研究组大鼠子宫内膜中MMP-9表达水平较模型组显著升高(P<0.05),而与对照组无显著差异(P>0.05)。结论藏药韦色尼阿丸预防大鼠子宫形成粘连有一定疗效,其作用机制可能与抑制子宫内膜组织TGF-β的表达及上调MMP-9的表达相关。

Abstract:Objective:To explore the effect of preventing intrauterine adhesions by Tibetan medicine Weisenia pill,and the influence on the expression levels of transforming growth factor-β(TGF-β)and matrix metalloproteinase-9(MMP-9)in endometrium.Methods:The female SD rats aged 6-8 weeks and weighing 180-200 g were randomly divided into 4 groups:normal control group,model control group,model group,study group.The rats in normal control group had no treatment;the rats in model control group were not modeled after uterine incision;the rats in model group were treated with the mechanical injury and alcohol burning method,while the rats in study group were treated with Weisenia pill for 14 days after model establishment.HE staining was used to observe and compare the changes of endometrial morphology and the number of endometrial glands between the groups.Masson staining was used to compare the area of endometrial fibrosis.The immunohistochemical staining was used to compare the effect of Tibetan medicine Weisenia pill on the expression of TGF-βand MMP-9.Results:Compared with the model group,the endometrial thickness and the number of glands significantly increased in the study group(P<0.05),but there were no significant changes in the endometrial thickness and the number of glands compared with the normal control group(P>0.05).The area of endometrial fibrosis in the study group was significantly less than that in the model group(P<0.05),but there was no significant change compared with the control group(P>0.05).The expression level of TGF-βin the endometrium of the study group was significantly lower than that of the model group(P<0.05),but there was no significant difference between the study group and the control groups(P>0.05).The expression level of MMP-9 in the endometrium of the study group was significantly higher than that of the model group(P<0.05),but there was no significant difference between the study group and the control groups(P>0.05).Conclusions:Weisenia pill,a Tibetan medicine,has a certain effect on preventing adhesion of uterus in rats.Its mechanism may be related to inhibiting the expression of TGF-βand up-regulating the expression of MMP-9 in endometrium.
