不同提取方法对黄芪多糖提取率影响的实验研究

摘    要:目的:考察用不同的提取方法(水提醇沉法、醇提后水提法、碱水提取法、碱醇提取法)提取对黄芪多糖含量的影响。方法:采用传统的苯酚-硫酸法,以葡萄糖为标准品,用紫外分光光度法建立多糖的含量测定方法,测定比较不同提取方法对黄芪多糖提取的影响。结果:得到葡萄糖的回归方程为:y=7.4978x-0.1488(r=0.9996),在0.024~0.120mg/mL内线性关系良好,平均回收率97.49%。不同提取方法提取率分别为:碱醇提取法提取率9.74%,碱水提取法提取率7.64%,醇提后水提法提取率3.62%,水提醇沉法提取率2.81%。结论:4种提取方法中碱醇提取法提取率最高,其次是碱水提取法,醇提后水提法,水提醇沉法提取率最低。

Abstract:Objective : To examine the effect of Astragalus Polysaccharides extraction rate with four different methods of extracting( extraction with water, extraction with water after alcohol, extraction with lye, extraction with alcohol base) Methods : The traditional methods of Phenol - Sulfuric acid were used, taking the glucose as the standard and UV was used to establish the corresponding standard curve. The effect of Astragalus Polysaccharides with four different methods of extracting was determined and compared. Results : The regression equation for glucose was : y = 7. 4978x - 0. 1488 ( r = 0.9996), with a good linear relationship in 0. 024 - 0. 120 mg/mL, and the RSD of precision was 2.21% ( n = 5 ), indi- cating the precision was good. The RSD of recovery was 1.68% (n = 5 ), the RSD of reproducibility was 2.10% (n = 6), indicating that the recovery and reproducibility for this experimental were good. The extraction rates of the method were as follows : Extraction with alcohol base was 9.74% , extraction with lye was 7.64% , extraction with water after alcohol was 3.62%, extraction with water was 2.81%. Conclusion:The results show that in these four methods, the highest rate of extraction is extraction with alcohol base, next is extraction with lye, extraction with water after alcohol and the lowest rate of extraction is extraction with water.
