康莱特联合光动力治疗胰腺癌及其机制的实验研究

摘    要:目的通过研究胰腺癌的细胞凋亡和细胞增殖,探讨康莱特注射液与光动力疗法(PDT)联用对人胰腺癌裸鼠皮下移植瘤的实验治疗的机制。方法通过把人胰腺癌细胞SW1990的组织块移植至裸鼠皮下而制备皮下移植瘤模型。60只胰腺癌模型随机分为6组:A组(荷瘤对照组,不予任何治疗);B组(1.25g/kg康莱特,照射前1d开始注射,连续10d);C组(2.5g/k康莱特,照射前1d开始注射,连续10d);D组(Photosan2mg/kg,腹腔内注射,48h后予激光照射);E组(B+D组);F组(C+D组)。每组10只。14d后,处死裸鼠,分别测量瘤体的大小及瘤重。通过免疫组化方法检测细胞凋亡和细胞增殖指数。结果康莱特注射液和光动力联用,对人胰腺癌SW1990的生长有很强的抑制作用,抑瘤率和细胞凋亡率高于单用疗法。细胞增殖指数则明显低于单用疗法。结论康莱特与光动力联用可诱导细胞凋亡,治疗效果明显优于单用疗法。

Experimental study of the therapeutic effect and mechanism of Kanglaite combined with photodynamic therapy on transplanted pancreatic cancer

Abstract:Objective To investigate the mechanism of Kanglaite injection (KI) combined with photodynamic therapy (PDT) on human pancreatic cancer xenograft in nude mice through the changes of apoptosis index (AI) and cell proliferation index (PI) . Methods The animal model of human pancreatic cancer was developed by suturing small pieces of SW 1990 tumors into the dorsum of nude mice. 60 animal models were randomly divided into six groups: A group (Control group, without any treatment), B group (receiving 1.25 g/kg KI i. v. continuously for 10 days), C group (receiving 2. 5 g/kg KI i. v. continuously for 10 days), D group (Photodynamic group, 2 mg/kg Photosan + laser irradiatiion), E group (B group + D group), F group ( C group + D group) . Each group involves 10 mice. Mice were killed on 14 days after treatment. Tumor weight and size were measured. The AI and PI of tumor were analysed by immunohistochemistry stain. Results KI combined with PDT had a significant inhibiory effect on tumor growth of pancreatic carcinoma. The tumor-inhibition rate and AI were higher than that of KI or PDT alone. PI were lower than that of KI or PDT alone. Conclusion KI combined with photodynamie therapy can induce apoptosis of tumor and has synergistic effect.
