消肿止痛合剂对大鼠急性脊髓损伤的影响

摘    要:目的:观察消肿止痛合剂对大鼠急性脊髓损伤的保护作用。方法:采用胥少汀脊髓半横切法制作大鼠T8-T9脊髓损伤模型,根据性别、体质量将60只大鼠随机分为6组,空白组(A),消肿止痛合剂低、中、高剂量组(B、C、D),甲基强的松龙组(E),模型组(F),每组10只,B、C、D组子消肿止痛合剂,每日用药量分别按成人等效剂量的5、10、20倍,每日用药3次,术后第8天开始改为每日2次,剂量不变;E组于术后开始肌注甲基强的松龙(0.03g/kg);A组和F组分别给予等量生理盐水。给药14天后,用放免法检测血清IL-6、NOS含量;免疫组化法测定Caspasc-3的表达;观察各组大鼠HE染色脊髓损伤组织的病理变化。结果:HE染色提示脊髓损伤组织改善情况D组优于C组,且与E组相似;损伤脊髓组织中IL-6、NOS含量F组显著增高,D、C组与E组相比有显著性差异(P〈0.01);Caspasc-3的表达在F组中升高,C组与E组、D组相比Caspasc-3表达有统计学意义(P〈0.01)。结论:消肿止痛合剂能降低IL-6、NOS在损伤脊髓组织中的含量,可抑制caspase-3在急性脊髓损伤后的表达。

Abstract:Objective: To observe the influence of XiaoZhong ZhiTong mixture on rats with acute spinal cord injury. Method: Rat models with T8 -T9 spinal cord injury were established with XU Shao -ring' s method of hemi-sectioned spinal cord injury. 60 rats were randomized into blank group (A), group of XiaoZhong ZhiTong mixture low-dose, medium-dose, high-dose (B, C, D), methylprednisolone group (E) and model group (F) on average based on gender and physique of the rats. Group of B, C, and D were administered with XiaoZhong ZhiTong mixture of 1/20, 1/10 and 1/5 of adult equivalent dosage, three times per day, then changed to twice per day at the eighth day after the operation; E group were given with intramuscular injection of methylprednisolone(30 mg/kg). Groups of A and F were given by same amounts of saline. After administration for 14 days, the contents of serum IL-6 and NOS were detected with radioimmunoassay; the expression of Capase-3 was detected with immunohistochemical method. Pathological changes of injured spinal tissues, stained with HE staining, in different groups were observed. Result: HE staining suggested that the improvement of injured spinal cord tissue in D group was superior to that in C group and similar to E group; The contents of IL-6 and NOS in injured spinal cord tissue in F group significantly raised, D and C group compared with E group showed significant difference (P〈0.01). The expression of Caspase-3 in F group raised, there was statistical meaning of the expression of Caspase-3 in C group compared with E group and D group (P〈0.01). Conclusion: XiaoZhong ZhiTong mixture can lower the level of IL-6 and NOS in the injured spinal cord tissue and inhibit the expression of caspase-3 after acute spinal cord injury.
