高效液相色谱法对绿茶中茶多酚含量的测定

摘    要:采用高效液相色谱法测定绿茶中茶多酚的含量。在色谱柱为Kromasil C18、流动相为乙酸:甲醇:N,N- 二甲基甲酰胺:水=1:1:40:58(V/V) 、色谱柱温30℃、进样量10μL、检测器为紫外检测器、检测波长280.0nm 的色谱条件下对绿茶中茶多酚含量进行测定。结果表明:回归方程为Y=1 × 10-7X - 0.0036(r=0.9995),方法变异系数小于3.04%,回收率为99.16%~100.60%;6 种绿茶样品茶多酚含量为62.07~110.86mg/g,说明产地不同,绿茶中茶多酚的含量相差较大。本方法简便、准确,适合绿茶中茶多酚含量的分析。

Abstract:The polyphenol contents in green tea were determined by high performance liquid chromatography method, using the mobile phase consisting of acetic acid, methyl alcohol, N,N- dimethylformamide and warter (1:1:40:58, V/V). The column temperature was 30 ℃, and the detection wavelength was 280.0 nm. The regression equation obtained was Y=1×10-7X-0.0036 (r =0.9995). The coefficient of variation was less than 3.04%, and the recovery was 99.16%-100.60%. The result indicates that the proposed method is simple , accurate and feasible for determination of polyphenols in the green teas.
