摘    要:用放射配基结合分析法测定了经腹腔注射重组人肿瘤坏死因子α型(TNFα)致伤的无菌大鼠脑细胞膜肾上腺素能β受体,同时用ELISA法测定了血中TNFα水平,用生化法测定脑细胞组织膜磷脂酶A_2(PLA_2)活性。结果表明,外原性重组人的TNFα在整体状况下能增加脑组织β受体数量,激活膜PLA_2,同时血中TNFα水平亦高于对照组。

Abstract:The effect of intraperitoneal tumour necrosis factor (TNF) on beta-adrenergic receptor Bmax and Kd as well as phospholipase A2 activity of brain tissue in germ free rats was studied. TNF group animals (n = 9) were given rhTNF (100ug/kg.d-1) intraperitoneally for three days, while to the control group animals (n = 9) 0.9% saline was given instead of rhTNF. All animals were sacrificed after last injection of rhTNF, and beta receptors and PLA2 activity in the brain tissue were assayed with radio-ligand binding assay and biochemical methods, while the serum TNF were determined by ELISA with the monoclone antibody of rat against rhTNFa. It was found that beta receptor Bmax and affinity to 3H-DHA were .significantly increased after the administration of non-lethel quantity of rhTNF (P<0.01), accompanied by activation of PLA2 (P<0.01), while the level of serum TNF was elevated about 7 times over that of the control group (P<0.01). The result suggested that exogenous rhTNF given to germ-free rats may regulate the function of beta-adrenergic receptor by activation of PLA2 in the brain tissue.
