大孔树脂纯化核桃楸皮总黄酮工艺

摘    要:目的大孔树脂纯化核桃楸皮总黄酮的工艺优化。方法以大孔树脂D-101、AB-8、NKA-2和HPD-826对总黄酮的吸附率和解吸率为指标筛选树脂种类,并对优选树脂的吸附特性和各影响因素进行研究,优化工艺条件。结果 AB-8具有较好的吸附率和解吸率,最佳纯化工艺为上样液的pH为7.0,总黄酮质量浓度为4.26 g·L(-1),上样流速为0.9 mL·min(-1),上样流速为0.9 mL·min(-1),洗脱溶剂为体积分数为50%的乙醇,洗脱体积为108 mL。经AB-8大孔树脂纯化1次后,核桃楸皮总黄酮含量提高至原来的3.12倍。结论 AB-8大孔树脂能较好地用于核桃楸皮总黄酮的纯化。

Abstract:Objective To study the purification technology of total flavonoids from Juglandis Mandshuricae Cortex using macroporous adsorptive resins. Methods The adsorption and desorption capacity of four types of resins (D-101, AB-8, NKA-2, and HPD-826) for total flavonoids was investigated to select the optimal macroporous resin for our purpose. The adsorption characters and the influencing factors of the selected macroporous resin were also investigated to optimize the purification technology. Results AB-8 macroporous resin had the best property for the purification of total flavonoids. The optimized conditions were as follows: the pH of extracted solution was adjusted to 7, the concentration of total flavonoids was 4.26 g·L-1, the flow rate was set at 0.9 mL·min-1, the elution reagent used was 50%(φ) ethanol, and the volume of elution was 108 mL. Under the optimized purification condition, the content of total flavonoids was increased by 3.12 fold as compared with the original extraction. Conclusion AB-8 macroporous resin is suitable to refine total flavonoids from Juglandis Mandshuricae Cortex.
