桑叶消渴胶囊对Ⅱ型糖尿病大鼠的影响

摘    要:目的 观察桑叶消渴胶囊对Ⅱ型糖尿病大鼠口服3 g/kg淀粉的糖耐量和连续给药29 d血糖、甘油三酯的影响.方法 将SPF级Wistar大鼠用链尿佐菌素(STZ)造成Ⅱ型糖尿病模型,测定甘油三酯.按照血糖随机分成空白对照组给予生理盐水、受试物低、高剂量组250 mg/kg、1 000 mg/kg,阳性对照二甲双胍200 mg/kg,糖耐量实验阳性对照改为拜糖萍20 mg/kg,ig量按10.0 ml/kg BW计算.糖耐量实验当天动物禁食过夜测定空腹血糖,给药时同时给予淀粉3 g/kg,于30 min、60 min、120 min后测血糖,并计算血糖升高值.降糖实验当天动物禁食4h后测定餐前血糖再给药,给药后240 min测定餐后血糖,尾静脉取血分离血清测定甘油三酯,计算各组给药后较给药前血糖下降值及血糖下降百分率.结果 受试物低、高剂量组能抑制Ⅱ型糖尿病大鼠糖吸收,与空白对照组比较60 min、120 min差异有统计学意义(P<0.05,P<0.01);连续给药29 d能降低Ⅱ型糖尿病大鼠的血糖,高剂量组与空白组比较差异有统计学意义(P<0.05);低、高剂量组与空白组比较均能降低甘油三酯,差异有统计学意义(P<0.05).结论 桑叶消渴胶囊剂量达到250 mg/kg对Ⅱ型糖尿病大鼠口服淀粉糖耐量有显著的抑制糖吸收作用,连续给药29 d剂量达到1 000 mg/kg能显著降低血糖和甘油三酯.

Abstract:Objective To observe the effect of Sangye Xiaoke Jiaonang(mulberry-leaf capsules) on the glucose tolerance test(GTT) in mice with type Ⅱ diabetes after oral intake of 3 g/kg amylum,and its effects on blood glucose and triglycerides after being administrated for 30 consecutive days.Methods Wistar rats(SPF) model with Type II diabetes was built by injection with streptozotocin(STZ).The blood and triglyceride was determined.The rats were randomized into a control group(saline),low-dose group(250 mg/kg mulberry-leaf capsules),high-dose group(1000 mg/kg mulberry-leaf capsules) and positive control group(200 mg/kg metformin).In GTT,the positive control group is Glucobay(20 mg/kg).The dose of administration is determined by 10.0 ml/kg BW.The fasting glucose was measured on the rats who had fasted over night on the day of GTT,The blood glucose was measured 30 min,60 min,120 min after intake of 3 g/kg amylum with mulberry-leaf capsules,with the growth of blood glucose level calculated.The rats fasted for 4 h were measured for the pre-dinner blood glucose levels before administration,and then detected for the post-dinner blood glucose levels and triglyceride levels 240 min after administration.The value and the rate of decline in blood glucose were analyzed.Results Glucose absorption was inhibited in the low-dose group and high-dose group,showing significant difference with that in the control group at 60 min and 120 min after administration(P<0.05,P<0.01).The blood glucose value in the model rats was declined after being administrated for 29 consecutive days.The difference between the high-dose group and the control group was statistically significant.The blood and triglyceride level was declined in low-dose group and high-dose group,showing statistically significant difference with the control group(P<0.05).Conclusion After being administrated mulberry-leaf capsules for 29 consecutive days at the concentration of 1 000 mg/kg,significantly declined levels of blood glucose and triglyceride could be determined in rats with type II diabetes,
