雌激素对谷氨酸诱导大鼠脑皮质细胞凋亡的影响

摘    要:目的:观察雌激素对谷氨酸诱导离体皮层神经元凋亡的作用,探讨雌激素的神经保护作用机制。方法:原代培养大鼠大脑皮质细胞,分为正常对照组、谷氨酸组和谷氨酸+雌激素组,采用TUNEL和免疫细胞化学检测等方法,分析雌激素对谷氨酸诱导神经元凋亡的作用。结果:谷氨酸可明显诱导神经元凋亡,凋亡指数为34.6%;雌激素能明显拮抗谷氨酸的凋亡毒性作用,凋亡指数下降为18.4%(与谷氨酸组相比P〈0.05);免疫组化结果显示,雌激素组bcl-2表达阳性细胞明显增加,bax蛋白表达受到抑制。结论:雌激素具有神经保护作用,作用机制可能通过影响凋亡相关基因的表达来实现。
