普鲁卡因对人肝癌HepG2细胞增殖影响的实验研究

摘    要:目的:探讨普鲁卡因(procaine)对体外人肝癌HepG2细胞增殖的影响。方法:应用不同浓度(0-10)mmol/L普鲁卡因处理HepG2细胞,倒置显微镜观察细胞形态学改变、四甲基偶氮唑盐比色法(MTT)检测细胞存活率、流式细胞术(FCM)观察细胞周期变化情况。结果:倒置显微镜观察普鲁卡因处理后的HepG2细胞呈现缩小、空泡、脱壁等形态学特征。MTT结果分析表明普鲁卡因能显著抑制HepG2细胞增殖,其抑制率呈药物浓度及时间依赖性。FCM结果显示普鲁卡因可以使HepG2细胞的G0/G1延长,S期缩短。结论:盐酸普鲁卡因能够使HepG2细胞的生长阻滞在G0/G1期,从而显著抑制HepG2细胞的增殖,并且具有剂量、时间依赖效应。

Abstract:Objective: To investigate the inhibitive effecs of Procaine on the human liver carcinoma HepG2 cells.Methods: The cell morphology was examined under a contrast microscope,cell proliferation was detected by MTT assay and cell cycles were examined by flow cytometry(FCM).Results: HepG2 cells showed shrinking,vacuoles,abscission.FCM showed that PCA prolonged G0/G1 phase and shortened the S phase of HepG2 cells.With the increase of PCA concentration and incubation time,HepG2 cells were inhibited increasingly in the proliferation determined by MTT.The effects on cell growth and proliferation depend on the concentration of PCA and incubation time.Conclusion: Procaine blocks the HepG2 cells in G0/G1 phase,and therefore inhibits the cell proliferation significantly in a dose-and time-dependent manner.
