肾透明细胞癌组织中miRNA-185水平的变化及其意义

目的探讨肾透明细胞癌组织中微小RNA-185（miRNA-185）水平的变化及其意义。方法提取22例手术患者肾癌组织和癌旁正常组织中的miRNAs,用RT-PCR的方法初步检测miRNA-185在肾癌组织和癌旁正常组织中的表达情况。采用Real-time PCR的方法定量分析miRNA-185在肾癌组织及癌旁正常组织中的表达情况。结果电泳结果显示,miRNA-185和U6 RNA均呈单一条带,其所处的位置及其在正常组织和癌组织中的表达情况符合前期的预计。Real-time PCR结果显示,miRNA-185在肾癌组组织中呈高表达,较癌旁组组织平均升高5.14倍。22例标本中有17例（77%）肾癌组组织中miRNA-185阳性表达率为77.3%（17/22）,明显高于癌旁组的22.7%（5/22）（P〈0.05）。G1期的4例肾癌标本均较相应的癌旁正常组织呈现低表达,高表达的则离散分布在G2和G3期。相关性分析结果显示,miRNA-185在肾透明细胞癌组织中的相对表达量与肾癌病理学分期呈正相关（r=0.58,P〈0.05）。结论 miRNA-185在肾癌组织中呈显著高表达,提示其与肾透明细胞癌的发生与演进有关,有望成为指导肾透明细胞癌诊断、治疗、预后的新靶点。

Change in miRNA-185 Expression Levels in Clear Cell Renal Cell Carcinoma and Its Significance

Objective To explore the change in miRNA-185 expression levels in clear cell renal cell carcinoma and its significance.Methods The miRNAs were isolated from kidney cancer tissue and adjacent normal tissue in 22 patients with clear cell renal cell carcinoma who had been operated on in recent years.Before using quantitative Real-time PCR to detect the expression of miRNA-185 in both normal and abnormal tissue,RT-PCR was performed as an initial test.ResultsElectrophoresis showed that both of miRNA-185 and U6 RNA gave a single band,and their position and expression in paratumor tissue and tumor tissue fit the expectation.Real-time PCR analysis showed that the expression of miRNA-185 in tumor tissue increased by 5.14 times compared with paratumor tissue.The positive expression rate of miRNA-185 in clear cell renal cell carcinoma was 77.3%（17/22）,which was obviously higher than that in paratumor tissue（22.7%,5/22）（P0.05）.The expression of miRNA-185 in G1 phase（4 samples） was higher than that in G2 and G3.Correlation analysis showed that the levels of miRNA-185 expression in human clear cell renal cell carcinoma was positively correlated with the pathology stage of renal carcinoma（r=0.58,P0.05）.Conclusion miRNA-185 is over-expressed in clear cell renal cell carcinoma,suggesting that it may be a new target for diagnosis,therapy and prognosis of clear cell renal cell carcinoma.
