| 摘 要： | 目的观察依硫磷酸对人慢性髓细胞白血病K562细胞系增殖的抑制作用。方法将依硫磷酸（粉末）以1640培养液稀释成0.2mg/ml、0.5mg/ml、0.75mg/ml、1.0mg/ml、1.6mg/ml、3.2mg/ml共6个浓度组,分别处理增殖期人慢性髓细胞白血病K562细胞,镜下观察不同时间点各浓度组细胞生长情况,应用CCK-8法检测细胞增殖能力,绘制生长曲线,计算抑制率和半数抑制浓度（IC50）。结果依硫磷酸对K562细胞增殖具有抑制作用,细胞倍增时间为48h,并且具有时间和剂量依赖性。在给药48h〉0.75mg/ml浓度的依硫磷酸对K562细胞增殖具有抑制作用（P〈0.05）,而3.2mg/ml浓度的依硫磷酸与K562细胞培养72h后对其增殖抑制作用最强,抑制率为91%。结论依硫磷酸对K562细胞增殖具有抑制作用,为临床应用依硫磷酸治疗慢性髓细胞白血病提供了实验基础。 |
