脐血造血干细胞分离方法的比较

摘    要:目的:探讨分离脐血造血干细胞的最佳方法。方法:采用Ficol分层法、3%明胶自然沉降法及6%羟乙基淀粉自然沉降法。结果:3%明胶自然沉降法所获有核细胞(NC)密度高达94.60(±23.80)×105/ml,CFUGM数为64.30(±12.18)/105NC,CD34+细胞数为1.21(±0.37)×105/ml,均较其它两种方法为高(P<0.05)。结论:3%明胶自然沉降法是一种较理想的分离脐血造血干细胞的方法。

Abstract:Objective: To find out an optimal method of separating hematopoietic stem cells (HSC) from umbilical cord blood. Methods: Density gradient centrifugation(FicollHypaque),3% gelatin natural sedimentation and 6% hydroxyethyl starch natural sedimentation were compared. Results: The number of nucleate cells,CFUGM and CD34 cells were 94.60(23.80)105/ml,64.30(12.18)/105 NC and 1.21(0.37)105/ml respectively by 3% gelatin natural sedimentation.These results were better than that from the other two methods (P<0.05). Conclusion: Three per cent gelatin natural sedimentation is an optimal umethod for separating HSC from umbilical cord blood.
