rhG-CSF前体脂质体的制备及体外释药动力学研究

摘    要: 目的制备重组人粒细胞集落刺激因子(rhG-CSF)脂质体并考察其理化性质和体外释放规律。方法采用前体脂质体法制备rhG-CSF脂质体,测定Zeta电位、粒度分布和包封率,比较rhG-CSF注射液和脂质体体内外释放情况。结果脂质体的平均粒径(195.2±40.8)nm,Zeta电位-(45.5±2.97)mV,包封率(84.2±0.36)%,体外释放具有明显的缓释特点。结论采用前体脂质体法制备的rhG-CSF脂质体,包封率较高,体内应用可显著延长半衰期。

Abstract:OBJECTIVE To study the preparation of rhG-CSF liposomes and its properties in vitro.METHODS The liposomes were prepared by pro-liposomes technique.The Zeta potential,particle size distribution,the entrapment efficiency and the release behavior of rhG-CSF liposomes in vitro were studied.RESULTS The mean diameters of rhG-CSF liposomes were(195.2±40.8) nm.The Zeta potential was-(45.5±2.97)mV.The mean entrapment efficiency was(84.2±0.36)%.The release of rhG-CSF in rhG-CSF liposomes were significantly delayed.CONCLUSION The rhG-CSF liposomes were prepared by pro-liposomes technique,with high entrapment efficiency and prolonged biological half life.
