摘    要:目的建立测定门冬氨酸含量的异硫氰酸苯酯柱前衍生化方法。方法采用高效液相色谱法测定门冬氨酸的含量。色谱柱为Phe—nomenexGeminiC18柱(250mmX4.6mm,5μm),流动相A为12.5mmol/L磷酸钠溶液(用磷酸调pH至6.8)、B为乙腈-水(80:20),梯度洗脱,流速为1.0mL/min,检测波长为254nm。结果门冬氨酸质量浓度在50.0~800.0μg/mL范围内与峰面积呈良好的线性关系(r=0.9998),平均回收率为99.18%,RSD=1.74%(n=9)。结论该方法准确、专属性强,稳定且无干扰,可用于门冬氨酸的含量测定。

Determination of Aspartic Acid with precolumn derivatization

Abstract:Objective To establish a high performance liquid chroatography(HPLC) method with precolumn derivatization for the deter- mination of aspartic acid content. Method The HPLC method was conducted on the Phenomenex Gemini C~8 column(250 mm x4. 6 mm, 5 ~m) with the mobile phase A of 12. 5 mmol/L sodium phosphate solution (adjusting pH to 6.8 with phosphoric acid) and the mobile phase B of aeetonitrile-water (80:20),the gradient elution was used. The flow rate was 1.0 mL/min;the detection wavelength was 254 nm. Results The mass concentration of aspartic acid in the range of 50. 0- 800.0 ~xg/mL had good linear relationship with the peak area (r = 0. 999 8),the average recovery rate was 99. 87%, RSD = 1. 741% (n = 9). Conclusion This method is accurate with strong specificity,stability and no interference, which be used for the determination of aspartic acid.
