响应面法优化超声辅助提取金丝皇菊多糖工艺及生理活性研究

摘    要:以金丝皇菊为试验材料,采用响应面分析法优化超声辅助提取金丝皇菊多糖的工艺,在单因素试验的基础上,以多糖提取率为响应值,依据Box-Behnken中心组合试验设计建立数学模型,优化金丝皇菊多糖的提取工艺。通过测定对O2-·、·OH、DPPH和ABTS自由基的清除作用评价其抗氧化活性,并初步探究温度和光照对其稳定性的影响。结果表明,金丝皇菊多糖最佳提取工艺为提取温度70℃、超声功率102W、乙醇浓度90%,多糖提取率为84.06mg/g,与预测值83.76mg/g相符。金丝皇菊多糖具有较好的抗氧化活性,当浓度为0.1mg/mL时,对DPPH、·OH、O2-·和ABTS自由基的清除率分别为64.73%、53.24%、51.93%、55.69%,IC50值分别为0.061、0.095、0.098和0.091mg/mL,且金丝皇菊多糖在常温(20℃)及避光条件下保留率最高,表现出较好的稳定性。

Abstract:Using Dendranthema morifolium(Ramat.)Tzvel.as experimental material,response surface analysis was used to optimize the technology of ultrasound-assisted extraction of polysaccharides from Dendranthema morifolium.Response surface analysis was used to optimize the process of ultrasound-assisted extraction of polysaccharides from Dendranthema morifolium.On the basis of single factor experiment,taking the extraction rate of polysaccharides as the response value,a mathematical model was established according to the design of Box-Behnken central combination experiment to optimize the extraction process of polysaccharides from Dendranthema morifolium.Its antioxidant activity was evaluated by measuring its scavenging effect on O2-·,·OH,DPPH and ABTS free radicals,and the effects of temperature and light on its stability were preliminarily explored.The results showed that the best extraction process of polysaccharide from Dendranthema morifolium was as follows:extraction temperature 70℃,ultrasonic power 102W,ethanol concentration 90%,and the extraction rate of polysaccharide was 84.06mg/g,which was consistent with the predicted value 83.76mg/g.Dendranthema morifolium polysaccharides have good antioxidant activity.When the concentration is 0.1mg/mL,the scavenging rates of DPPH,·OH,O2-·and ABTS free radicals are 64.73%,53.24%,51.93%and 55.69%,respectively,and the IC50 values are 0.061,0.095,0.098 and 0.091mg/mL,respectively.The retention rate of Dendranthema morifolium polysaccharides is the highest at room temperature(20℃)and under the condition of avoiding light,showing good stability.
