香菇多酚超声波提取工艺及抗氧化性分析

摘    要:以干香菇为原料,乙醇溶液作为提取溶剂,在单因素试验的基础上,结合响应面法优化了香菇多酚的提取工艺,并采用大孔树脂吸附法对香菇多酚进行分离纯化,通过体外试验来评价其抗氧化活性。试验结果表明,香菇多酚的最佳工艺参数为乙醇浓度50%、提取温度74℃、料液比1∶42(g/mL)、超声功率146 W,在此条件下,香菇多酚得率为10.91 mg/mL,分离纯化后得到的50%乙醇洗脱物中的多酚含量最高,达到41.89%。抗氧化活性表明,香菇多酚对DPPH自由基、ABTS+自由基和羟自由基具有明显的清除能力,其香菇多酚粗提物的抗氧化能力低于香菇多酚纯化物。

Abstract:Using dry Lentinus edodes as the raw material and ethanol solution as the extraction solvent,on the basis of single factor experiment,the extraction technology of polyphenols from Lentinus edodes is optimized combined with response surface methodology,and the macroporous resin adsorption method is used to separate and purify polyphenols from Lentinus edodes,and its antioxidant activity is evaluated by in vitro experiment.The results show that the optimum technological parameters of polyphenols from Lentinus edodes are ethanol concentration 50%,extraction temperature 74℃,solid-to-liquid ratio 1∶42(g/mL)and ultrasonic power 146 W.Under these conditions,the yield of polyphenols from Lentinus edodes is 10.91 mg/mL,and the content of polyphenols in 50%ethanol eluent is the highest of 41.89%after separation and purification.The antioxidant activity shows that polyphenols from Lentinus edodes have obvious scavenging ability on DPPH free radical,ABTS free radical and hydroxyl free radical,and the antioxidant ability of crude extracts of polyphenols from Lentinus edodes is lower than that of purified extracts.
