血液稀释联合局部亚低温对大鼠全脑缺血/再灌注损伤的影响

摘    要:目的观察血液稀释联合局部亚低温对大鼠全脑缺血/再灌注损伤的影响。方法 40只雄性SD大鼠随机分为缺血/再灌注损伤组(I/S)、局部亚低温组(MH)、急性等容性血液稀释组(NH)和局部亚低温联合急性等容性血液稀释组(MHNH),每组10只。血液稀释后3h处死大鼠,采用ELISA法测定血清S-100B的浓度,采用干湿重法测定大脑半球的含水量,应用电镜观察脑皮质神经元超微结构。结果与I/S组比较,其他3组大鼠血清S-100B浓度、脑含水量明显下降(P<0.05),MHNH组最低,皮质神经元损伤最轻。结论急性等容性血液稀释联合局部亚低温后大鼠血清S-100B的浓度明显下降,对缺血/再灌注损伤大鼠有保护作用。

Abstract:Objective To investigate the effects of hemodilution combined with local mild hypothermia on brain ischemia-reperfusion injury in rats.Methods Forty adult male Sprague-Dawley rats were randomly assigned to 4 groups:ischemia-reperfusion group(I/R group),mild hypothermia groups(MH group),acute normovolemic hemodilution(NH) and mild hypothermia combined with acute normovolemic hemodilution group(MHNH group),10 rats in each group.The rats were killed 3 h after hemodilution.The concentration of serum S-100B was assessed by ELISA at the end of the study.Brain water content wag was determined by dry wet weight method.The ultrastructure changes of cerebral cells of rat were analyzed with electron microscopy.Results Compared with group I/R,the serum S-100B protein levels and brain water content of other groups were significantly decreased(P < 0.05),in which MHNH group was the lowest.MHNH group The severity of injury of the cortex neurons in rats was ameliorated effectively.Conclusion Hemodilution combined with local mild hypothermia can decrease concentration of serum S-I00B induced by ischemia-reperfusion injury in rats and can protect brain against I/R injury.
