荷瘤裸鼠口腔鳞状细胞癌乏氧状况的初步研究

摘    要:目的检测荷口腔鳞状细胞癌裸鼠肿瘤组织乏氧区域分布,明确厌氧菌能否在肿瘤乏氧区内生长,为认识和开发利用肿瘤乏氧区域提供依据。方法荷瘤裸鼠尾静脉注射^99mTc-HL91,应用SPECT检测鳞癌组织及正常组织乏氧区域分布。荷瘤裸鼠尾静脉注射青春双歧杆菌菌悬液,切取鳞癌组织及其它正常组织,匀浆后厌氧培养。结果^99mTc-HL91在肿瘤组织内高浓度聚集,与肿瘤体积呈线性相关;肿瘤区域、肿瘤对侧部位及头胸部位平均像素^99mTc-HL91吸收值有显著性差异。肿瘤组织内可见青春双歧杆菌生长,其它正常组织内未见青春双歧杆菌生长。结论口腔鳞癌组织中乏氧区域分布广泛,且与肿瘤体积密切相关。口腔鳞癌组织乏氧微环境适合严格厌氧菌-青春双歧杆菌的生长。

A preliminary study of hypoxia status in mice with oral squamous cell carcinoma

Abstract:Objective To investigate the distribution of hypoxia in oral squamous cell carcinoma of tumor bearing nude rats and to determine if the microenvironment of hypoxia is suitable for the growth of Bifidobacterium Adolescentis (BA). Methods Mice bearing oral squamous cell carcinoma were injected with ^99mTc-HL91 and then scanned by SPECT. Region of interest (ROI) technique was used to determine the positive imaging interested region and the negative imaging interested region in the same mice. The correlation between ^99mTc-HL91 uptake density and tumor volume was analyzed. Mice bearing oral squamous cell carcinoma were injected with Bifidobacterium Adolescentis and then normal tissues and tumor tissues were excised and minced respectively. A sample of each tissue was cultured in TPY media for 3 days. Results All mice were hypoxia-positive,with the uptake of ^99mTc-HL91 in the tumor tissue being significantly higher than in normal tissue. The uptake density of ^99mTc-HL91 increased with the size of the tumor. Bifidobacterium Adolescentis was found in the tumor tissue after 3 days of cultivation in TPY media, and the number increased with the size of the tumor. There was no BA found in normal tissues. Conclusion There are broad hypoxic areas in the oral squamous cell carcinoma tissues, and the microenvironment of hypoxia is suitable for growth of the strictly anaerobic bacterium Bifidobacterium Adolescents.
