氯胺酮对大鼠缺血/再灌注心肌NF-κB和肿瘤坏死因子的影响

为观察氯胺酮对缺血 /再灌注心肌 NF-κB和血清肿瘤坏死因子 (TNF-α)的影响 ,将健康 SD大鼠 2 4只随机分为心包打开假手术组 (A组 )、缺血 /再灌注对照 (I/ R)组 (B组 )、5 mg· kg- 1氯胺酮 (KTM) + I/ R组 (C组 )和 10 mg·kg- 1 KTM+ I/ R组 (D组 )。在缺血 3 0 min,再灌 12 0 min后 ,取心肌组织用 Western-blot法分别检测其中胞核 NF-κB含量。在再灌 3 0 min和 12 0 min两个时间点分别取颈静脉血清做 TNF-α的 ELISA检测。结果显示 ,B、C、D组胞核 NF-κB含量均高于 A组 ,升高程度呈下降趋势 (P<0 .0 1) ,且两个时间点血清 TNF-α浓度也高于 A组 ,升高程度也呈下降趋势 (P<0 .0 1或 P<0 .0 5 )

Effects of ketamine on myocardial expression of NF-kB and serum TNF-α level in ischemia-reperfused rats

To explore the effects and possible mechanism of ketamine on myocardial expression of NF κB and serum TNF αlevel in ischemia reperfused(I/R)rats model. 24 healthy SD male rats were divided randomly into control group (A group), I/R group(B group), 5mg·kg -1 ketamine+I/R group (C group), 10mg·kg -1 ketamine+I/R group(D group). After ischemia of half an hour and reperfusion of two hour, the myocardium was exampled to examine their neronal cellular NF κB expression. When reperfusion for 30 minutes and 120 minutes, serum was exampled to measure their TNF αlevel. The expression of serum TNF αin two points and myocardial neronal NF κB of group B、C、D were higher than group A, But the level of increasing in B、C、D. was descending respectively ( P <0.01 or P <0.05).
