当归对大鼠肺纤维化间质成纤维细胞的干预作用

摘    要:目的:观察浓当归对博莱霉素诱导大鼠肺纤维化程度的影响及对肺间质成纤维细胞的影响。方法:45只Wistar大鼠随机分为空白组、模型组和当归组,经气管内灌注博来霉素后,当归组大鼠腹腔内注射浓当归注射液10 ml/(kg.d),空白组、模型组给予等量生理盐水处理,分别于第7,14和28天各处死5只大鼠。免疫组化法分析各组α-平滑肌动蛋白(-αSMA)、胶原蛋白Ⅰ(COL-Ⅰ)、胶原蛋白Ⅲ(COL-Ⅲ)表达水平,用原位杂交的方法检测结缔组织生长因子(CTGF)mRNA水平。结果:当归组大鼠肺组织中COL-Ⅰ、COL-Ⅲ沉积较模型组明显降低(P<0.05);当归组间质中-αSMA阳性表达水平较模型组明显减低(P<0.05),当归组大鼠肺组织CTGF mRNA水平较模型组降低(P<0.05)。结论:当归可能通过降低纤维化肺组织中CTGF mRNA水平,间接抑制成纤维细胞增殖、分化作用,减轻纤维化程度。

Abstract:Objective: To investigate the effect of angelica sinensis on bleomycin induced pulmonary fibrosis in rats.Methods: Forty-five Wistar rats were divided into control group,model group and angelica group randomly.After intratracheally instilled with bleomycin,the angelica group were treated with angelica sinensis injection 10 ml /kg·d intraperitoneally,while the control group and model group were treated with the same volume of normal saline until they were killed.On day 7,14 and 28 after instillation,seven rats of each group were sacrificed.Immunohistochemical method was used to detect the expression of α-SMA,collagenⅠ and collagenⅢ protein.The level of CTGF mRNA was observed by in situ hybridization method.Results: The collagenⅠand collagenⅢ content in pulmonary tissue of angelica group were remarkably decreased than those of model group in every stage(all P<0.05).The CTGF mRNA level and the α-SMA expression level of angelica group was lower than that of model group respectively(both P<0.05).Conclusion: Angelica sinensis might alleviate bloemycin induced pulmonarry fibrosis in rats,and inhibit lung interstial α-SMA positive myofibroblast through decreasing the CTGF mRNA expression.
