条浒苔多糖的超声波辅助提取及其抗氧化活性

摘    要:为了开发利用绿潮藻类浒苔中含量丰富的碳水化合物,以多糖提取率为指标,首先通过单因素试验确定提取条件,然后采用均匀设计优化超声波辅助提取工艺条件。采用普鲁士蓝反应及Fenton体系测定了浒苔多糖的总还原力及对羟自由基的清除活性。结果表明:浒苔多糖的热水提取条件为料液比1∶20,pH 9.0,温度80℃,时间6h;超声波辅助提取条件为总时间10min、超声时间5.5s、间隙时间5.5s、总体积82.5mL、功率712.5W,超声波处理后热水提取6h,在此条件下多糖提取率为17.64%~18.84%。浒苔粗多糖具有一定还原力和较强的羟自由基清除作用,EC50分别为32.79mg/mL和4.10mg/mL。

Abstract:In order to utilize the carbohydrate in Enteromorpha clathrata, a type of green tide al- ga, the extraction process of polysaccharides from it was investigated using uniform design and its antioxidant activity was tested. Results show that the optimal hot water extracting conditions is as follows: solid to liquid 1:20, pH 9.0, temperature 80 ℃, and the extracting time 6 h. The ul- trasonic-assistant extraction condition is as follows: the total ultrasonic time 10 rain, ultrasonic time 5.5 s, interval time 5.5 s, total volume 82.5 mL, and the output power of ultrasonic 712.5 W. The sample was extracted by ultrasonic-assistant extraction under the above conditions, and the extraction rate was 17. 64%- 18. 84%. Antioxidant test revealed that the polysaccharides from E. clathrata showed certain reduction power and scavenging activity on hydroxyl free radi- cal. The EC50 of reduction power and scavenging activity on hydroxyl free radical was 32.79 mg/ mL and 4.10 mg/mL respectively.
