附子多糖对H22和S180荷瘤小鼠的抗肿瘤作用研究

摘    要:目的 :研究附子粗多糖和酸性多糖对S180、H2 2荷瘤小鼠的抑瘤作用并探讨其作用机制。方法 :采用水提醇沉法得到附子粗多糖和酸性多糖 ,通过腹腔注射和灌胃两种途径给药 ,以肿瘤生长抑制率和存活延长率为指标观察附子多糖对荷瘤小鼠 (S180和H2 2 )的抑瘤作用 ,并通过检测淋巴细胞转化能力、NK细胞活性、肿瘤细胞凋亡率、癌基因和细胞增殖指数 (PI)等指标探讨其抗肿瘤作用机制。结果 :附子粗多糖和酸性多糖对H2 2荷瘤小鼠肿瘤有显著的抑瘤作用 ,灌胃给药的抑瘤率分别为 4 5 30 %和 5 9 36 % (P <0 0 1) ,腹腔给药的抑瘤率分别为 4 9 6 5 %和 6 9 2 8% (P <0 0 1)。两种多糖对S180荷瘤小鼠肿瘤也有较显著的抑制作用。附子多糖对S180和H2 2荷瘤小鼠有延长存活时间的作用 (P <0 0 5或P <0 0 1)。两种多糖均明显增大了小鼠脾脏的重量 (P <0 0 1) ,提高了荷瘤小鼠的淋巴细胞转化能力 (P <0 0 5 )和NK细胞活性 (P <0 0 1) ,提高了抑癌基因p5 3和Fas的表达 (P <0 0 1) ,并且提高了肿瘤细胞凋亡率 (P <0 0 1)。结论 :附子粗多糖和酸性多糖有显著的抑瘤作用 ,其作用机制主要是增强机体的细胞免疫功能 ,诱导肿瘤细胞凋亡和调节癌基因的表达。

Abstract:Objective:To investigate the antineoplastic effect of crude Monkshood polysaccharide (CMP) and acid Monkshood polysaccharide (AMP) on mice transplanted with the tumor cell line H22 and S180.Methods:The AMP and CMP were obtained by water extraction and alcohol precipitation.The Kunming mice with H22 and S180 tumor were given peros (PO)or by intraperitoneal injection (IP).The inhibiting rate of tumor growth and life prolongation rate were obscrved,and the lymphocyte transformation capacity,the activity of natural killer(NK) cells,apoptotic rate of tumor tissue,proliferation index (PI) and oncogenes were tested by MTT,LDH method and flow cytometry respectively.Results:CMP and AMP could significantly inhibit the tumor growth.The inhibiting rate of tumor growth in H22(PO) was 45 30% (CMP) and 59 36%(AMP) and in H22{IP} was 49 65%(CMP) and 69 28% (AMP).There were significant differences between the control group and experimental group ( P <0 05 or P <0 01).The result on mice with S180 tumor was alike to that of H22.CMP and AMP could also notably prolong the survival of mice ( P <0 05 or P <0 01),enhanced the spleen weight of rice( P <0 01),increased lymphocyte transformation capacity and the activity of NK cells ( P <0 01 or P <0 05).The results of flow cytometry indicated that CMP and AMP could increase apoptotic rate obviously ( P <0 01),decrease PI and increase expression of p53 and Fas.Conclusion:CMP and AMP have obvious antineoplastic effect,the main mechanism is to enhance the cellular immunologic function,induce apoptosis of tumor tissue and regulate expressions of oncogens.
