摘    要:目的:从千山上一个温泉周围的土壤中分离、纯化出一株新耐热菌FR3,并对其各项特征性质进行研究。方法:菌株从千山上一个温泉周围的土壤中分离出来。菌液经培养、发酵涂布在平板上,50℃培养后,纯化菌株,选取个体形态和菌落形态都比较特殊的菌株FR3做进一步实验。分别对菌株的形态特征、生长特征及生理生化特征进行了具体研究。结果:分离出的菌株FR3为革兰氏阳性,在生长过程中细菌细胞的形态有球杆的变化,到恒定期变成类球形细胞。菌株FR3有芽孢、荚膜、鞭毛、运动性,无伴胞晶体,V.P.实验呈阴性,能降解淀粉,不能降解纤维素,兼性好氧,生长的pH值为5-11、温度30-60℃;最适生长的pH值为7.5、温度55℃。菌落初形成时是乳白色边缘整齐的圆形,后随机向3个方向发展,最终形成不规则圆形菌落、花瓣形菌落或者扁平扩散形菌落。结论:通过比较可以初步确定,菌株FR3是一株还没有被鉴定的新耐热菌。

Abstract:Objective:To research characters of a thermophilic bacterium FR3which isolated and purified from the soil around a hot spring in Qian mountains.Methods:After foster and ferment,bacterium was smear into the slab,Foster and purify bacterium again under 50 ℃.Then the bacterium FR3which have special individual characters was choosed to study.Its characteristic of morphology、growth and biology especially were researched.Results:The cells of isolate FR3 strain was Gram positively,the morphology of the cells of isolate was changed from rod to round in the process of growth,the cells in the initial phase of logarithmic growth were long rod,until to invariable phage of growth,the cells changed to globe.V.P.test was negative,could catabolism starch,could not catabolism fibrin,aerobic,but could grow without oxygen,grow at and between 30~60 ℃(op t.55℃),grow at and between pH 5~11(op t.pH 7.5).Conclusion:The bacterium FR3is a new thermophilic bacterium which is never be discovered.
