摘    要:目的:构建抗人卵巢癌全套单链抗体基因噬菌体表面呈现库。方法:利用噬菌体表面呈现技术,构建基因库。经过panning筛选富集后,用ELISA方法检验抗原结合活性。结果:从单个噬菌体克隆中筛选到26个具有抗原结合活性的阳性克隆。结论:噬菌体表面呈现技术为构建抗人卵巢癌全套单链抗体基因噬菌体表面呈现库提供了一条更为有效的新途径。

Construction and preliminary selection and identification of phage display library of anti-human ovarian cancer single chain Fv antibody
Abstract:AIM: To construct the phage display library of anti human ovarian cancer single chain Fv antibody. METHODS: The ScFv gene library was constructed by phage display technology. The affinity to antigen was detected by ELISA after panning. RESULTS: Twenty six antigen binding clones were identified from 94 individual phagemid clones. CONCLUSION: In the production of engineering antibodies against human cancer, this strategy may provide an alternative approach.
