日本血吸虫在小鼠体内不同发育时点的生长速度与蛋白组分差异

摘    要:目的比较观察日本血吸虫发育中虫体在不同时点的生长速度和蛋白组分差异。方法选择昆明鼠人工感染日本血吸虫尾蚴,分别收集感染前和感染后第8、12、16、20、24和28d虫体;用MoticBA400病理图像处理系统及3.2分析软件采集虫体图象作形态观察和大小测量(包括水平面积、周长、长与宽);用SDS-PAGE平行分离各时点虫体蛋白,用图像分析软件QuantityOne4.4比较分析其区带差异。结果日本血吸虫感染昆明鼠后,发育中虫体在不同时点的生长速度不均衡,第8~12天的生长速度为最快,其他依次为12~16d、16~20d、20~24d、0~8d和24~28d的虫体;同一时点虫体间发育的个体差异也大,以12d后虫体较为明显。电泳分析7个不同时点的虫体蛋白区带显示:区带数在0、8、12、16、20、24和28d虫体中分别为27、30、33、32、31、36和29条,其中多数为相同分子量蛋白区带,但有些在蛋白表达量上有明显差异;各时点虫体特异性区带在0、8、12~24d和28d虫体中分别为7、4、8、3条,虫体特异性区带多为低表达量的条带。结论日本血吸虫童虫在小鼠体内的不同发育时点间存在着生长速度和蛋白组分的差异,其中以8~12d时段的虫体发育最快,两者间蛋白组分差异也较明显。

Abstract:Objective] To observe and compare the differences of growth rate and protein fractions at different developmental time points of schistosoma japonicum. Methods] Worms were collected from cercariae infected Kunming strain of mice at different time points (0, 8, 12, 16, 20, 24 and 28 days post-infection), respectively. The morphology was observed and the areas, perimeters, lengths and widths measured with pathological Image Processing System Motic BA400 and 3.2 software. After prepared from S.j at different developmental time points, the individual soluble proteins were separated by SDS-PAGE and stained with kaomasi blue R-250. The band sizes and their differences were analyzed with quantity one 4.4. Results] After invading into the mice, the differences of growth velocities of worms at different developmental time points were conspicuous and disproportion. As evaluated the growth velocities with growth rate, worms from the day 8 to12 showed the fastest development trendlines, followed by worms from the day 12 to 24, and then worms from day 0 to 8. The worms from the day 24 to 28 revealed the slowest growth velocities. At the same development time point, there were some distinctions among the worms, and the distinction was more obvious after day 12. Electrophoreses analysis of worm proteins from 7 time points showed that. the numbers of protein bands in 0, 8,12,16, 20,24 and 28-day old worms were 27, 30, 33, 32, 31, 36 and 29, respectively. Most molecular weights of the protein fractions were identical, but there were manifest differences of express quantities in some identical main bands. Furthermore, there were 7, 4, 8, 3 characteristic bands of the polypide in 0, 8, 12 to 24 and 28 old worms, and most of the characteristic bands were low-expression ones. Conclusion] There were differences in the growth rates and protein components in different developmental time points of Schistosomula japonicum. Evaluated with growth rate, the growth velocity of 8 to 12-day old warms was the fastest among them. The difference of proteins fractions was more obvious between 8d and 12d worms.
