巴戟天醇提物对骨髓基质细胞增殖影响的血清药理学实验方法的建立

摘    要:探讨巴戟天醇提物对骨髓基质细胞增殖影响的血清药理学实验方法的建立。方法采用MTT法检测各组给药剂量、给药时间、采血时间、血清浓度的含巴戟天醇提物的血清培养大鼠BMSCs72小时后的OD值。结果等剂量组(给药剂量),4天组(给药时间),1小时组(采血时间)的OD值,与对照组比较有显著性差异(P<0.05),各组培养血清浓度的OD值20%组>10%组>5%组。结论巴戟天醇提物等剂量灌胃,给药4天,末次给药1小时后腹主动脉采血,制备血清后,以20%的浓度培养,为促进大鼠BMSCs增殖的优选方案。

Abstract:Objective:To study the foundation of serum pharmacology method on the effect of alcohol extract of Morinda officinalis to the proliferation of bone marrow stromal cells. Methods: According to the MTT method,to detect the OD value of every group of dose and time of fill rat's stomach,time of drawing out blood and thickness of serum after their blood serum had cultured rat bone marrow stromal cells for 72 hour. Results: The OD value of the same dose group,four days group and one hour group had notability difference comepare with the comparison group ( P <0.05). The order of OD value of every group's thickness of blood were 20%group>10%group>5%group. Conclusion:It was the most excellent project of accelerating the proliferation of rat bone marrow stromal cells that fill rat's stomach with the same dose alcohol extract of Morinda officinalis How and lasting out four days, drawing out blood on one hour after the last time fill rat's stomach, culturing bone marrow stromal cells with 20%thickness of serum.
