肝细胞生长因子对小鼠实验性结肠炎结肠细胞因子的影响

摘    要:背景:肝细胞生长因子(HGF)是一种多功能因子,近年文献报道其有可能成为炎症性肠病(IBD)的辅助治疗药物。目的:研究HGF对小鼠实验性结肠炎结肠细胞因子的影响,探讨HGF用于IBD辅助治疗的可能机制。方法:30只健康昆明小鼠随机分为正常对照组、结肠炎模型组和HGF干预组,后两组以嗯唑酮溶液灌肠诱导结肠炎模型。造模后予HGF干预组小鼠HGF100μg/d腹腔注射,连续4d。评估各组疾病活动指数(DAI),第13d处死所有小鼠,观察结肠大体和组织学损伤情况.以荧光定量聚合酶链反应(PCR)检测结肠黏膜白细胞介素(IL)-4、IL-5、IL-10、干扰素(IFN)-γ和肿瘤坏死因子(TNF)-αmRNA表达。结果:与正常对照组相比,结肠炎模型组的DAI和结肠大体、组织学损伤评分以及结肠黏膜IL-4、IL-5、IL-10、IFN-γ、TNF-αmRNA表达量显著增高(P〈0.01);HGF干预组上述指标均较结肠炎模型组显著降低(P〈O.01),与正常对照组相比无明显差异。结论:HGF可能通过抑制结肠黏膜细胞因子的表达,改善小鼠实验性结肠炎的结肠炎症反应。

Abstract:Background: Hepatocyte growth factor (HGF) is a muhifunctional factor. Recently, it was demonstrated to be a potential adjuvant drug in the treatment of inflammatory bowel disease (IBD). Aims: To investigate the effect of HGF on colonic cytokines in mice with experimental colitis, and the potential mechanism of HGF used as an adjuvant in the treatment of IBD. Methods: Thirty healthy Kunming mice were randomized into normal control group, colitis model group and HGF intervention group. In the latter two groups, colitis was induced by intrarectal administration of oxazolone. HGF 100 μg/d was injected intraperitoneally for 4 days after induction of colitis in HGF intervention group. Disease activity index (DAI) was assessed during the experiment. All mice were sacrificed on the 13th day and macroscopic and microscopic colonic damage were assessed. Expressions of interleukin (IL)-4, IL-5, IL-IO, interferon (IFN)-γ/and tumor necrosis factor (TNF)-α mRNA in colonic mucosa were determined by fluorescent quantitative polymerase chain reaction (PCR). Results: Compared with normal control group, DAI, macroscopic and microscopic colonic damage score, as well as expressions of IL-4, IL-5, IL-10, IFN-γ/and TNF-α mRNA in colonic mucosa increased significantly in colitis model group (P〈O.01). The above-mentioned indices in HGF intervention group were significantly lower than those in colitis model group (P〈0.01) and similar to those in normal control group. Conclusions: HGF can attenuate the colonic inflammation in mice with experimental colitis via inhibiting the expressions of cytokines in colonic mucosa.
