摘    要:目的:建立不同产地枇杷中齐墩果酸、熊果酸的含量测定方法。方法:采用RP—HPLC法,齐墩果酸、熊果酸采用乙醇超声提取30分钟,用Agilent Eclipse XDB—C18柱,甲醇-乙腈-0.05%醋酸铵溶液(12:67:21)为流动相,流速1.0ml/min,检测波长205nm。结果:齐墩果酸和熊果酸的线性范围分别为2.61—209mg.L^-1(r=0.9998),7.85—628mg.L^-1(r=0.9999),与峰面积呈良好线性关系,平均加样回收率依次为97.27%,99.75%。结论:本方法操作简单,结果准确,重现性好,适用于琵琶中齐墩果酸、熊果酸含量测定。

Determination of oleanolic acid and ursolic acid in branch and leaf of Eriobotrya Japonica by HPLC

Abstract:Objective: To establish a method for determination of oleanolic acid and ursolic acid in Eriobotrya Japonica from different habitats by HPLC. Method: Oleanolic acid and ursolic acid was extracted with ethanol solution by ultrasound for 30 minutes, and the content of berberine was determined by RP - HPLC. Agilent C18 column (250 mm ×4. 6 mm, 5 μm) was used for the determination, with a flow rate of 1. 0 mL · min - 1 and column temperature of 25℃. The mobile phase were methanol - acetonitrile - 0. 05% solution of ammonium acetate ( 12 : 67 : 21 ) and detection wave length was 205nm. Result: There was good linearity in the range of 2. 61 -209mg. L^-1 (r=0. 9998), 7. 85-628mg. L^-1 (r=0. 9999) ofoleanolic acid and ursolic acid. Average recovery rate was 97. 27%, 99. 75%. Conclusion: The method is simple, reliable and repeatable, and is suitable for the determination of oleanolic acid and ursolic acid in Eriobotrya Japonica.
