摘    要:目的 建立超高效液相色谱-串联质谱法测定野生蘑菇中3种鹅膏毒肽的方法。方法 样品经甲醇超声提取,氮吹干后,加水复溶,HLB 固相萃取柱净化,Hypersil Gold - C18色谱柱(150 mm×2.1 mm,1.9 μm)分离,电喷雾(ESI+)多反应监测(MRM)方式检测。结果 3种鹅膏毒肽在5.0~100 ng/ml范围具有良好线性关系,相关系数值均大于0.999,平均回收率和相对标准偏差分别在81.8%~91.8%和4.5%~9.3%,定量限低于30 μg/kg。结论 该方法能准确、灵敏测定野生蘑菇中的3种鹅膏毒肽,适用于野生蘑菇中鹅膏毒肽的检测。

Abstract:Objective Method for the determination of three kinds of amatoxins in wild mushrooms was established by ultra-performance liquid chromatography-tandem mass spectrometry in this paper. Methods Samples were firstly extracted with methanol by ultrasonic extraction, then nitrogen drying, water redissolution, HLB solid phase extraction column purifying and Hypersil Gold-C18 column(150mm×2.1mm, 1.9μm) separation, lastly detected by electrospray(ESI+) in multiple-reaction monitoring(MRM) mode. Results There was a good linear relationship for the amatoxins determination in the range of concentration 5.0 to 100 ng/mL with the correlation coefficient value greater than 0.999. The average recoveries and relative standard deviations were between 81.8% to 91.8% and 4.5% to 9.3%, respectively. Quantification limit was less than 30 μg/kg. Conclusion The method can accurately and sensitively determine the three kinds of Amanita peptides in wild mushrooms, which is suitable for the detection of Anoxis peptides in wild mushrooms.
