外周血内皮祖细胞移植对肺动脉高压兔模型肺动脉压力及其结构的影响

摘    要:目的探讨外周血内皮祖细胞(EPCs)移植对野百合碱导致的兔模型肺动脉高压的治疗效果。方法选取健康新西兰兔50只,随机将其分为3组:(1)正常对照组(n=16);(2)野百合碱诱导肺动脉高压组(MCT模型组,n=16);(3)EPCs治疗组(n=18)。EPCs治疗组兔行外周血单个核细胞成功诱导、分化为内皮祖细胞之后,将其从兔尾静脉注入进行移植,正常对照组及MCT模型组注射相同剂量生理盐水3周后对3组肺动脉压、右心室肥大指数以及肺血管重构的情况进行观察与对比。结果与正常对照组比较,MCT模型组肺动脉压升高(30.13 mm Hg±3.12 mm Hg vs.15.99 mm Hg±2.23 mmHg),右心室肥大指数增大(0.51±0.06 vs.0.26±0.03),肺动脉管壁厚度增加(28.77μm±4.35μm vs.8.44μm±0.05μm),肺小动脉厚度增加(73.78μm±7.61μm vs.40.16μm±2.17μm),差异均有统计学意义(P<0.05)。与MCT模型组比较,EPCs治疗组各项指标均显著改善,肺动脉压为(22.78±2.71)mm Hg,右心室肥大指数为(0.41±0.05),肺动脉管壁厚度为(16.55±3.15)μm,肺小动脉厚度为(53.50±3.77)μm,差异均有统计学意义(P<0.05)。结论外周血内皮祖细胞移植治疗兔模型肺动脉高压,可在一定程度上修复损伤的肺血管系统,有利于改善肺动脉高压。

Abstract:Objective To study therapeutic effect of peripheral blood endothelial progenitor cells(EPCs) transplantation on pulmonary arterial hypertension in rabbit model induced by monociotaline.Methods 50 healthy New Zealand rabbits were selected and randomly divided into 3 groups:(1)normal control group(n =16),(2)pulmonary arterial hypertension group induced by monocrotaline(MCT model group,n =16),(3) EPCs treatment group(n =18).Rabbits peripheral blood mononuclear cells were induced and differentiated into EPCs,then injected into the tail vein of rabbit in EPCs treatment group,same volume of normal saline were injected in normal control group and MCT model group.After 3 weeks,pulmonary artery pressure,right ventricular hypertrophy index and pulmonary vascular remodeling were observed and compared.Results Compared with normal control group,pulmonary artery pressure of MCT model group increased(30.13 mm Hg±3.12 mm Hg vs.15.99 Hg mm±2.23 mm Hg),right ventricular hypertrophy index increased(0.51±0.06 vs.0.26±0.03),pulmonary artery wall thickness increased(28.77 μm±4.35 μm vs.8.44 μm±0.05μm),thickness of pulmonary arterioles increased(73.78 μm±7.61 μm vs.40.16 μm±2.17 μm),the differences were statistically significant(P〈0.05).Compared with the MCT model group,all indexes of EPCs treatment group were significantly improved,pulmonary arterial pressure was(22.78±2.71) mmHg,right ventricular hypertrophy index was(0.41±0.05),pulmonary artery wall thickness was(16.55±3.15)μm,pulmonary artery thickness was(53.50±3.77) μm,the difference were statistically significant(P〈0.05).Conclusion The transplantation of peripheral blood endothelial progenitor cells could affect pulmonary artery hypertension in rabbit model,repair the lung vascular system damage to a certain extent,improve the pulmonary arterial hypertension.
