乌司他丁对大鼠胰腺缺血再灌注损伤bcl-2和HSP70表达的影响

摘    要:目的探讨乌司他丁对大鼠胰腺缺血再灌注损伤的保护作用及其机制。方法SD大鼠72只随机分为假手术组、缺血再灌注组和乌司他丁组。采用免疫组化方法分别检测胰腺缺血30min再灌注0h、6h、12h及24h时胰腺组织中bcl-2蛋白和热休克蛋白70(HSP70)表达的变化,同时观察胰腺组织病理学改变。结果乌司他丁组在再灌注6h、12h、24h时bcl-2蛋白表达明显高于缺血再灌注组和假手术组(P〈0.05),后两组各时点表达水平比较无显著差异(P〉0.05);缺血再灌注组和鸟司他丁组HSP70的表达在12h、24h较假手术组明显增加(P〈0.05)。病理组织学观察:乌司他丁组炎症反应较缺血再灌注组明显减轻。结论乌司他丁对大鼠胰腺缺血再灌注损伤有明显保护作用,其机制可能与上调胰腺bcl-2蛋白表达有关。

Effects of ulinastatin on the expression of bcl-2 and HSP70 proteins in ischemia reperfusion injury of rat pancreas

Abstract:Objective To study the effect of ulinastatin during ischemia - reperfusion injury of rat pancreas and assess the possible mechanism. Methods Seventy two SD rats were randomly divided into sham operation group, ischemia - reperfusion group and ulinastatin group. Expression of bcl - 2, HSP70 in pancreas ceils and pancreas histopathology were observed after 30 minutes of pancreas ischemia and reperfusion for 0 h, 6 h, 12 h and 24 h respectively. Results The expression level of bcl - 2 in ulinastatin group was obviously higher than that of sham operation group and ischemia - reperfusion group at 6 h, 12 h and 24 h of reperfusion( P 〈 0.05 ). The expression level of HSP70 in ischemia - reperfusion group and ulinastatin group was higher than that in sham operation group at 12 h and 24 h of reperfusion( P 〈0.05) ; Histopathologic observation indicated that the inflammation was alleviated in ulinastatin group. Conclusion Ulinastatin has protective effects on rat pancreas ischemia - reperfusion injury. The protective effect of ulinastatin may be associated with the up - regulation of bcl - 2 expression.
