香菇菌丝原生质体制备及融合条件的研究

摘    要:讨论不同菌龄、酶解时间、酶解温度、渗透压稳定剂对香菇原生质体产率的影响。结果表明 ,香菇菌丝制备原生质体时最适菌龄为 6天 ,酶解时间为 3h ,酶解温度为 34℃ ,渗透压稳定剂用 0 .6mol/L甘露醇为最佳。在此基础上 ,以 35 %的PEG为融合诱导剂进行香菇B0 1、L2 6种间原生质体融合 ,并用显微镜观察到融合子的形成

Abstract:The influence of different cell age, enzymolysis time, temperature and osmotic pressure stabilizer on productive rate of protoplast from Lentinula edodes were discussed in this paper. It is the best conditions of preparing protoplast with Lentinula edodes hyphae that cell age was 6 days, That enzymolysis time was 3 hours, that enzymolysis temperature was 34℃ and that concentration of mannitol as osmotic pressure stabilizer was 0.6mol/l. In addition, the protoplast from Lentinula edodes B01and L26 were fused with 35% polyetlylene glycol (PEG) solution as fusion inducer and formation of fusant was observed under the microscope.
