HPLC法测定利福平在血清中的浓度并作老年患者的药代动力学研究

目的：以高效液相色谱法测定患者血清中利福平浓度，并对老年肺结核患者进行药代动力学研究。方法：10名＞65岁肺结核患者，口服利福平0.45 g，不同时间点采集其血样，用HPLC法测定血药浓度，用DAS 3.1软件计算药代动力学参数。结果与结论：该方法简单、灵敏、准确，适用于体内利福平药代动力学分析。利福平在老年患者与年轻人群的药代动力学特征存在较大差异。

Objective: To determine the concentration of rifampicin (RFP) in patient plasma by HPLC and investigate the pharmacokinetics of RFP in elder patients. Methods: A single 0.45 g dose of RFP was given to 10 tuberculosis patients over 65 years old by oral administration. Blood samples were preceded by protein precipitation and the plasma concentrations of RFP were determined by an HPLC method built for this study. Data were analyzed by the DAS3.1 software. Results and Conclusion: The HPLC method described in this report was of high sensitivity and selectivity for accurate determination of the plasma concentrations of RFP in human. The major pharmacokinetic parameters of RFP were calculated by using non compartmental model. The results showed that the pharmacokinetics of RFP in elder patients was significantly different from normal.
