摘    要:建立了高效液相色谱法测定发酵液中丙酮酸含量方法。采用氨基柱进行液相色谱检测,检测波长210nm,柱温30℃,流动相采用乙腈/磷酸二氢钾。对检测方法的检出限、定量限、线性、精密度、稳定性进行考察。该方法线性方程为:y=0.3246x+0.051,在5-200μg/mL内,线性关系良好(r=0.9999)。该方法适合于发酵液中丙酮酸含量测定。

Abstract:To develop a HPLC method for analysing pyruvate in pyruvic acid form entation broth was established. ZORBAX NH2 column was used and mobile phase was Acetonitrile/K2HPO3.The column temperature was 30 ℃ and the detection warelength was at 210 nm. Detection method for the detection limit, limit of quantification, linearity, precision and stability study. Results show that the method was linear over the concentration range of 5 to 200 μ g/mL, the value of correlation coefficiant(r)was 0.999 9. The method was successfully applied to determine the pyruvate in pyruvate fermentation broth.
