体外破骨细胞性骨吸收陷窝的动态观察

摘    要:动态观察破骨细胞的功能及药物的作用。采用体外破骨细胞培养法、显微摄影、显微光密度仪及图像分析等技术。结果表明,破骨细胞具有不规则的移行性,在一定时间范围内,随着培养时间的延长,破骨细胞性骨吸收陷窝不断增多与增大,并呈不规则形状。国产帕屈磷酸钠(APD)可直接抑制骨吸收陷窝的数目及表面积。提示动态观察体外破骨细胞性骨吸收陷窝形态变化的方法是一种直观、简便、稳定和能反映药物作用的方法。

Abstract:The function of osteoclasts and the roles of drugs were observed dynamically by means of culturing osteoclasts on bone slices, the techniques of photomicrography, microdensitometric scan and computer image analysis. The results indicated that osteoclasts could irregularly move along, and the number and range of osteoclastic resorbing lacunae increased with the incubation period until 112 hours. APD made in China reduced the number and surface area of osteoclastic bone resorbing lacunae. It suggests that the method is objective, steady and convenient to reflect the activity of drugs.
