摘    要:目的:体外诱导和扩增感染伯氏疟原虫小鼠骨髓来源的树突状细胞(DC),并探讨其与疟疾发病机制的关系。方法:感染伯氏疟原虫的BALB/c实验小鼠组与正常对照组小鼠各7只,将其股骨骨髓细胞分离。去除悬浮细胞,贴壁细胞中加入细胞因子(IL-4和GM-CSF)继续进行培养,2周后,诱导的DC经相差显微镜进行观察。结果:DC在体外经细胞因子的刺激可增殖,但实验组小鼠DC增殖速度和数量明显低于对照组小鼠(P<0.05)。结论:感染伯氏疟原虫的小鼠DC形态和数量的改变,可能由此导致其功能的改变,这可能是造成疟疾患者感染持续发展的原因之一。

Abstract:Objective:To in vitro induce and proliferate dendritic cells(DCs) from the bone marrow of mice infected by Boshi Plasmodium and explore the relationship between DCs and malaria pathogenesis.Methods:The bone marrow cells were flushed from the femurs of 7 mice infected by Boshi Plasmodium and 7 healthy mice of control group,respectively.The floating cells were removed and the adherent cells continued to be cultured with cytokines(IL-4 and GM-CSF).Two weeks later,the induced DCs were observed with the phase microscope.Results:DCs were proliferated by cytokine stimulation in vitro,and the DCs proliferation ability and the number of the experiment group mice were significantly decreased than those of the control group mice(P<0.05).Conclusion:The changes in the DCs morphology and number could cause the function change,which is one of possible reasons why malaria patients keep on development.
