基于大鼠血浆中盐酸小檗碱药代动力学前处理方法研究

摘    要:目的:探讨灌服以盐酸小檗碱为主要成分的药物在大鼠血浆中最佳的前处理方法。方法:采用均匀设计试验法对血浆样品前处理中溶剂浓度和加入量进行考查,优选出最佳血样处理条件;以盐酸小檗碱为考察指标,采用高效液相色谱法测定其在血样中的含量。色谱柱:Agilent C18(150 mm×4.6 mm,5μm);流动相:乙腈-水(40∶60)(每100 mL含0.34 g磷酸二氢钾,含0.17 g十二烷基磺酸钠);流速:1.0 mL/min,检测波长:349 nm;柱温:30℃。结果:盐酸小檗碱的最佳血样处理方法:甲醇加入量为血样量体积的4倍,甲醇浓度为100%;盐酸小檗碱的回收率达93.02%,RSD为2.31%。结论:盐酸小檗碱的最佳血样处理方法稳定可行,为以盐酸小檗碱为主要入血成分的药物在体内的药动学的研究奠定了基础。

Abstract:Objective: To observe the best treatment method for extraction of berberine hydroehloride by gastric perfusion. Methods : The best treatment method for rat blood was selected by using Uniform Design Experimentation. The content of berberine hydrochloride was chosen as the index and HPLC method was used to determine the berberine hydrochloride content. Chromatogram column: AgilentC18(150 mm× 4.6 mm,5 × m); the mobile phase: Acetonitrile-aqueous solution(40:60)(per 100 mL by adding 0.34 g KH2PO4 and 0.17 g SDS); the flow rate:1.0 mL/min; the UV detector was operated at 349 nm and the column temperature was controlled at 30 ℃ .ResuIts : The best processing method was 4 times volume of 100% methanol processing blood samples. The reeovery rate of berberine hydrochloride reached 93.02% and RSD was 2.31%. Conclusion : The best treatment method for extraction of berherine hydroehloride is stable. To give priority to berberine hydrochloride drugs in vivo pharmacokinetic study laid a foundation.
