灰树花子实体中水溶性多糖提取工艺优化研究

摘    要:以水为浸提液,通过单因素试验研究了颗粒度、浸提温度、浸提时间、水料比、醇沉度等因素对灰树花子实体多糖提取率的影响,并采用正交试验对提取工艺进行优化。试验表明,水料比对灰树花子实体多糖提取率的影响最大,其次是浸提时间,浸提温度影响最小。通过对提取条件的优化,结合收益、成本等综合因素选出最佳优化工艺为:浸提温度90℃,浸提时间5 h,水料比25∶1。验证试验显示,在最佳工艺条件下提取的多糖提取率达13.4%。

Abstract:The aim is to optimize the extraction technology of polysaccharides from Grifola frondosa.With the water as the extraction solvent,the effects of particle size,extraction temperature,extraction time,solid-liquid ratio,degree of alcohol precipitation on the yield of polysaccharides from Grifola frondosa are investigated by the single factor experiments and the extraction technology is optimized by using the orthogonal experiment.The results show that the effect of solid liquid radio extraction temperature on the yield of polysaccharides from Grifola frondosa is the largest,the second is the effect of extraction time and the extraction temperature is the least.The optimization technology suitable for local conditions which is selected through optimizing extraction condition and combining the comprehensive factors of benefit,cost and so on is as follows: the extraction temperature is 90 ℃,the extraction time is 5 h and the liquid solid radio is 25∶1.The verification test shows that the yield of polysaccharides extracted under the optimum technology condition is 13.4%.
